Abstract

This disclosure relates to novel morphinan compounds and their derivatives, pharmaceutically acceptable salts, solvates, and hydrates thereof. This disclosure also provides compositions comprising a compound of this disclosure and the use of such compositions in methods of treating diseases and conditions that are beneficially treated by administering a σ₁ receptor agonist that also has NMDA antagonist activity.

Claims

1. A method for treating diabetic neuropathic pain comprising administering to a subject in need thereof a therapeutically effective amount of a compound of formula I:

2. The method of claim 1, wherein R¹ is CD₃ and R² is CH₃.

3. The method of claim 1, wherein R¹ is CD₃, and R² is not CH₃.

4. The method of claim 1, wherein any atom not designated as deuterium is present at its natural isotopic abundance.

5. The method of claim 1, comprising administering a compound of Formula I or a pharmaceutically acceptable salt thereof with a second therapeutic agent selected from quinidine, quinidine sulfate, oxycodone, and gabapentin.

6. The method of claim 5, wherein the second therapeutic agent is quinidine sulfate.

7. The method of claim 5, wherein the dosage amount of the compound of Formula I or pharmaceutically acceptable salt thereof is 10-60 mg, and the dosage amount of quinidine is 2.5-30 mg.

8. The method of claim 2, wherein any atom not designated as deuterium is present at its natural isotopic abundance.

9. The method of claim 3, wherein any atom not designated as deuterium is present at its natural isotopic abundance.

10. The method of claim 5, wherein in the compound of formula I or pharmaceutically acceptable salt thereof R¹ is CD₃, and R² is CH₃.

11. The method of claim 5, wherein in the compound of formula I or pharmaceutically acceptable salt thereof R¹ is CD₃ and R² is not CH₃.
