Abstract

Devices, systems, and methods are disclosed herein for treatment of a disease, disorder, or condition in a vertebrate subject. A device is provided that includes one or more passive cooling elements configured to be applied to one or more tissues of a vertebrate subject to modulate at least one activity of brown adipose tissue of the vertebrate subject, wherein at least a portion of the one or more passive cooling elements is configured to be implantable, and wherein the one or more passive cooling elements includes a first portion configured to be in association with the one or more tissues to be cooled, and a second portion configured to be placed in association with one or more tissues adjacent to an external epidermal tissue of the vertebrate subject.

Claims

1. A method for modulating an activity of brown adipose tissue in a vertebrate subject comprising:

inducing non-shivering thermogenesis in brown adipose tissue by applying cooling to a pulmonary vein and to one or more brown adipose tissues of the vertebrate subject with an implantable cooling system having at least a first portion including one or more cooling elements and a second portion including one or more cooling elements, the first portion at least partially implantable proximate a pulmonary vein and the second portion at least partially implantable proximate one or more brown adipose tissues;

wherein inducing non-shivering thermogenesis in the brown adipose tissue includes

activating at least the first portion implantable proximate the pulmonary vein so as to induce activation of thermoreceptors associated with the pulmonary vein, and

activating the second portion implantable proximate the one or more brown adipose tissues so as to induce non-shivering thermogenesis of the brown adipose tissue.

2. The method of claim 1, wherein the second portion further includes one or more branches configured to be placed in association with the one or more brown adipose tissues adjacent to the external epidermal tissue of the vertebrate subject.

3. The method of claim 1, wherein inducing non-shivering thermogenesis in brown adipose tissue further includes applying cooling to one or more thermoresponsive tissue of the vertebrate subject with at least one cooling element from the first portion.

4. The method of claim 1, wherein inducing non-shivering thermogenesis in brown adipose tissue further includes applying cooling to one or more nervous tissue of the vertebrate subject with at least one cooling element from the first portion.

5. The method of claim 1, wherein inducing non-shivering thermogenesis in brown adipose tissue further includes applying cooling to one or more cutaneous tissue of the vertebrate subject with at least one cooling element from the first portion.

6. The method of claim 1, wherein inducing non-shivering thermogenesis in brown adipose tissue further includes applying cooling to one or more tissues in a core of a body of the vertebrate subject.

7. The method of claim 6, wherein inducing non-shivering thermogenesis in brown adipose tissue further includes applying cooling to one or more blood vessels or lymph vessels.

8. The method of claim 1, further comprising controlling the one or more cooling elements of the first portion or the second portion with at least one programmable controller operably configured to provide instructions to the one or more cooling elements of the first portion or the second portion in response to information regarding one or more physiological conditions of the vertebrate subject.

9. The method of claim 1, further comprising sensing with one or more sensors information regarding one or more physiological conditions of the vertebrate subject and communicating the information from the one or more sensors to at least one programmable controller.

10. The method of claim 9, further comprising comparing the information regarding the one or more physiological conditions to information of a standard value or preprogrammed value.

11. The method of claim 9, further comprising activating one or more pumps with the at least one programmable controller to control flow through one or more cooling elements of the first portion or the second portion.

12. The method of claim 9, further comprising receiving information regarding one or more physiological conditions from an outside operating source via a receiver including at least one of the programmable controller or a digital processing unit.

13. The method of claim 12, wherein receiving the information regarding one or more physiological conditions from the outside operating source includes receiving information regarding one or more physiological conditions from a computing device or a human operator.

14. The method of claim 9, wherein receiving the information regarding the one or more physiological conditions includes receiving information regarding a plasma level of one or more metabolic analytes in the vertebrate subject.

15. The method of claim 9, wherein receiving the information regarding the one or more physiological conditions includes receiving information regarding a sugar level or a fatty acid level in the vertebrate subject.

16. The method of claim 1, further comprising activating, via at least one programmable controller, one or more pumps to control flow through the one or more cooling elements of the first portion or the second portion.

17. The method of claim 1, wherein the one or more cooling elements of the first portion or the second portion includes one or more heat pumps.

18. The method of claim 17, wherein the one or more heat pumps transfer heat from one or more tissues to be cooled including blood or skin of the vertebrate subject.

19. The method of claim 1, wherein one or more cooling elements of the first portion or the second portion includes one or more implantable deep tissue heat-extracting components in combination with one or more surface tissue heat-releasing components.

20. The method of claim 1, further comprising applying cooling to a lumen of the vertebrate subject.

21. The method of claim 20, further comprising applying cooling to a circulatory vessel of the vertebrate subject.

22. The method of claim 20, further comprising applying cooling to a colon or a large intestine of the vertebrate subject.

23. The method of claim 1, wherein applying cooling to the one or more brown adipose tissues of the vertebrate includes cooling the one or more brown adipose tissues with one or more cooling elements configured to cool by one or more chemical cooling elements.

24. The method of claim 1, wherein applying cooling to the one or more brown adipose tissues of the vertebrate includes cooling the one or more brown adipose tissues with one or more cooling elements of the first portion to attain a brown adipose tissue temperature from approximately 4°

C. to approximately 36°

C.

25. The method of claim 1, wherein applying cooling to the one or more brown adipose tissues of the vertebrate includes cooling the one or more brown adipose tissues with one or more cooling elements to attain a brown adipose tissue temperature from approximately 12°

C. to approximately 20°

C.

26. The method of claim 1, wherein applying cooling to the one or more brown adipose tissues of the vertebrate includes cooling the tissue with one or more cooling elements to attain a brown adipose tissue temperature from approximately 24°

C. to approximately 32°

C.

27. The method of claim 1, further comprising cooling the tissue with the one or more cooling elements to attain a brown adipose tissue temperature approximately 16°

C. or lower.

28. The method of claim 1, further comprising providing a power source.

29. The method of claim 28, wherein the power source includes stored power, a battery, a fuel cell or beamed power.

30. The method of claim 1, further comprising providing one or more medicaments for treatment of weight loss, metabolic disorder, diabetes, obesity, metabolic syndrome, dyslipidemia, or hypercholesterolemia, configured to be applied to the one or more brown adipose tissues in combination with the one or more cooling elements of the first portion.

31. The method of claim 30, wherein the one or more medicaments include one or more of β

-adrenergic receptor agonist, NPY antagonist, leptin, UCP activating agent, thyroxine, serotonin reuptake inhibitor, MCH antagonist, GLP-1 agonist, 5-HT2c agonist, 5-HT2A agonist, galanin antagonist, CRF agonist, urocortin agonist, melanocortin agonist or enterostatin agonist.

32. The method of claim 1, further comprising activating a neurostimulator proximate the one or more brown adipose tissues.

33. The method of claim 1, further comprising activating a neurostimulator activating one or more second tissues of the vertebrate subject.

34. The method of claim 33, wherein the one or more second tissues include nerve tissue.

35. The method of claim 1, further comprising activating at least one of electric neurostimulator, magnetic neurostimulator, ultrasonic neurostimulator, or microwave neurostimulator proximate the one or more brown adipose tissues.

36. A method for inducing weight loss in a vertebrate subject comprising:

detecting via one or more sensor a glucose level of the vertebrate subject; and

inducing non-shivering thermogenesis in brown adipose tissue by applying cooling to one or more brown adipose tissues and to a pulmonary vein of the vertebrate subject with an implantable cooling system having at least a first portion including one or more cooling elements, and a second portion including one or more cooling elements, the first portion at least partially implantable proximate one or more brown adipose tissues, the second portion at least partially implantable proximate a pulmonary vein;

wherein inducing non-shivering thermogenesis in the brown adipose tissue includes activating the first portion implantable proximate the one or more brown adipose tissues so as to induce non-shivering thermogenesis of the brown adipose tissue, and activating at least the second portion implantable proximate the pulmonary vein so as to induce activation of thermoreceptors associated with the pulmonary vein, responsive to a detected target glucose level.

37. The method of claim 36, wherein applying cooling to the one or more brown adipose tissues with the implantable cooling system includes applying cooling to the one or more brown adipose tissues via one or more branches configured to be placed in association with the one or more brown adipose tissues adjacent to the external epidermal tissue of the vertebrate subject.

38. The method of claim 36, further comprising activating one or more cooling elements of the first portion or the second portion via at least one programmable controller operably configured to provide instructions to the one or more cooling elements of the first portion or the second portion in response to information regarding one or more physiological conditions of the vertebrate subject.

39. The method of claim 36, further comprising sensing with one or more sensors information regarding one or more physiological conditions and communicating the information from the one or more sensors to at least one programmable controller.

40. The method of claim 39, further comprising comparing the information regarding the one or more physiological conditions to information of a standard value or preprogrammed value.

41. The method of claim 39, further comprising activating one or more pumps with the at least one programmable controller to control flow through one or more cooling elements of the first portion or the second portion.

42. The method of claim 36, further comprising activating one or more cooling elements of the first portion or the second portion in response to receiving information regarding one or more physiological conditions from an outside operating source via a receiver including at least one of the programmable controller or a digital processing unit.

43. The method of claim 36, wherein applying cooling to the one or more brown adipose tissues and to the pulmonary vein of the vertebrate subject with an implantable cooling system includes applying cooling sufficient to increase metabolic activity or increase proliferation of the one or more brown adipose tissues in the vertebrate subject.
