Abstract

The invention provides novel derivatives of cyclopamine having the following formula:

1. A compound represented by the following structure:

2. The compound of claim 1, wherein R¹ is H, —

OR, amino, sulfonamido, sulfamido, —

OC(O)R⁵ or —

N(R⁵)C(O)R⁵.

3. The compound of claim 2, wherein R¹ is sulfonamido.

4. The compound of claim 1, wherein R² is H or 3-7 membered heterocycloalkyl that includes from 1-4 heteroatoms independently selected from nitrogen, oxygen, and sulfur.

5. The compound of claim 4, wherein R² is H.

6. The compound of claim 1, wherein R¹ and R² taken together along with the carbon to which they are bonded form ═

12. The compound of claim 1, wherein R¹ is sulfonamido, R² is H and R³ is H.

13. The compound of claim 1, wherein R¹ is sulfonamido, R³ is H and R⁴ is H.

14. The compound of claim 1, wherein R¹ is sulfonamido, R² is H and R⁴ is H.

15. The compound of claim 1, wherein R² is H, R³ is H and R⁴ is H.

16. The compound of claim 1, wherein said compound is isolated.

17. The compound of claim 1, wherein said compound is epimerically pure.

20. A pharmaceutical composition comprising a compound of claim 1, or a pharmaceutically acceptable salt thereof, and at least one pharmaceutically acceptable excipient.

24. The compound of claim 1, wherein each R⁵ is, independently, alkyl or aralkyl.
