An Outer Membrane Vesicle (OMV) from a gram-negative bacterium, wherein the OMV comprises at least one heterologous protein free in its lumen, and which OMVs are capable of eliciting an immune response against the heterologous protein when administered to a mammal, the heterologous protein is functionally active in the lumen of the OMV, the heterologous protein is a soluble protein, the heterologous protein is (a) a periplasmic protein in a heterologous organism and the native signal sequence is replaced by the signal sequence of the periplasmic protein, or (b) a membrane-associated protein in a heterologous organism and deletion of the membrane anchor, wherein the gram-negative bacterium is escherichia coli and wherein the heterologous protein is a double mutant of extracellular cholesterol-dependent streptolysin O (Slo-dm) from streptococcus pyogenes or the cell envelope serine protease SpyCep from streptococcus pyogenes.
