1. A droplet actuator comprisinga. A top substrate and a bottom substrate separated to form a droplet operations gap therebetween;

  • b. at least one set of reservoir electrodes disposed on the bottom substrate; and

    c. at least one on-actuator reservoir formed in the top substrate between the top substrate and the at least one set of reservoir electrodes on the bottom substrate, the at least one on-actuator reservoir comprising;

    i. a loading port comprising a cup for containing a quantity of liquid, the cup comprising an upper portion that is closed but includes an opening, wherein the cup is fitted over an upwardly projecting outlet portion of the loading port;

    a liquid storage area;

    a droplet metering zone; and

    a droplet distribution zone;

    wherein each of the storage, metering, and dispensing regions is characterized by a different gap height of the droplet operations gap between the top substrate and the bottom substrate.
