Abstract

The invention provides methods for producing a plant with altered resistance to powdery mildew, the methods comprising transformation of a plant with a genetic construct including a polynucleotide encoding of a polypeptide with the amino acid sequence of SEQ ID NO: 1 or a variant of fragment thereof. The invention also provides isolated polypeptides, polynucleotides, constructs and vectors useful for producing a plant cell and plants transformed to contain and express the polypeptides, polynucleotides and constructs. The invention also provides plants produced by methods of the invention.

Claims

1. An isolated polynucleotide encoding a polypeptide that comprises the sequence of SEQ ID NO:

1 or a fragment or variant thereof, wherein the fragment or variant confers resistance to powdery mildew in a plant.

2. The isolated polynucleotide of claim 1, wherein the fragment comprises a sequence with at least 70% identity to the sequence of SEQ ID NO:

3. The isolated polynucleotide of claim 1, wherein the fragment comprises a sequence with at least 70% identity to the sequence of SEQ ID NO:

39. An isolated polynucleotide comprising:

a) a polynucleotide comprising a fragment, of at least 15 nucleotides in length, of a polynucleotide of any one of claims 1 to 16 and 24 to 33;

b) a polynucleotide comprising a complement, of at least 15 nucleotides in length, of the polynucleotide of any one of claims 1 to 16 and 24 to 33;

or

c) a polynucleotide comprising a sequence, of at least 15 nucleotides in length, capable of hybridising to the polynucleotide of any one of claims 1 to 16 and 24 to 33 under stringent hybridisation conditions.

40. A genetic construct which comprises a polynucleotide of any one of claims 1 to 16, 24 to 33, and 39.

41. An expression construct which comprises a polynucleotide of any one of claims 1 to 16, 24 to 33, and 39.

42. An RNAi construct which comprises a polynucleotide of any one of claims 1 to 16, 24 to 33, and 39.

43. A host cell comprising the construct any one of claims 40 to 42.

44. A plant cell which comprises the construct of any one of claims 40 to 42.

45. A plant cell genetically modified to express a polynucleotide of any one of claims 1 to 16, 24 to 33, and 39, or a polypeptide of any one of claims 17 to 23 and 34 to 38.

46. A plant which comprises a plant cell of claim 44 or 45.

47. The plant of claim 46, which has increased resistance to powdery mildew.

48. A method for producing a plant cell or plant with increased resistance to powdery mildew, the method comprising transformation of a plant cell or plant with the polynucleotide of any one of claims 1 to 16, 24 to 33, and 39.

49. A method for selecting a plant with increased resistance to powdery mildew, the method comprising testing of a plant for altered expression of a polynucleotide of any one of claims 1 to 16, 24 to 33, and 39, or a polypeptide of any one of claims 17 to 23 and 34 to 38.

50. A group of plants selected by the method of claim 49.

51. A plant cell or plant produced by the method of claim 48.

52. A part, propagule or progeny of the plant of any one of claims 46, 47 and 51, wherein, the plant is genetically modified to include a polynucleotide of claims 1 to 16, 24 to 33, and 39, or polypeptide of any one of claims 17 to 23 and 34 to 38.

8. An isolated polynucleotide comprising the sequence of SEQ ID NO:
