Abstract

The present disclosure provides a nucleic-acid extraction method for carrying out the following procedures in the same cell, the procedures including: performing an ultrasonic process on a sample containing a nucleic acid; adsorbing substances contained in the sample by making use of an adsorption carrier; and condensing the nucleic acid by damming the nucleic acid moving in an electrophoresis phenomenon. In accordance with the nucleic-acid extraction method, it is possible to carry out an ultrasonic process, an adsorption process and an electrophoresis process in the same cell. Thus, it is possible to eliminate an operation to transfer the sample from one cell to another one for each of the processes.

Claims

1. A nucleic-acid extraction method for carrying out the following procedures in the same cell, the procedures comprising:

performing an ultrasonic process on a sample containing a nucleic acid;

adsorbing substances contained in said sample by making use of an adsorption carrier; and

condensing said nucleic acid by damming said nucleic acid moving in an electrophoresis phenomenon.

2. The nucleic-acid extraction method according to claim 1, wherein a cation exchange resin or a zeolite is used as said adsorption carrier.

3. The nucleic-acid extraction method according to claim 2, wherein a strongly acidic cation exchange resin is used as said cation exchange resin.

4. The nucleic-acid extraction method according to claim 3, wherein said ultrasonic process is carried out after blending bead particles in said sample.

5. The nucleic-acid extraction method according to claim 4, wherein said substances are each a foreign substance other than said nucleic acid.

6. The nucleic-acid extraction method according to claim 5,

wherein said procedure of performing said ultrasonic process on said sample is a procedure of performing said ultrasonic process on said sample after diluting said sample by making use of a buffer solution, and

the pH of said buffer solution has a value in a range of 4.0 to 8.0.

7. The nucleic-acid extraction method according to claim 6, wherein said buffer solution contains a thickening agent.

8. The nucleic-acid extraction method according to claim 7, wherein said thickening agent contains polyethylene glycol and/or hydroxy ethyl cellulose.

9. A nucleic-acid extraction cartridge comprising:

a channel allowing a sample containing a nucleic acid to be introduced into said channel;

electrodes each provided on one of the ends of said channel;

a damming section configured to divide said channel into a negative-electrode side area and a positive-electrode side area and dam said nucleic acid;

an adsorption carrier provided in said negative-electrode side area to adsorb substances contained in said sample; and

an ultrasonic-wave generation section configured to carry out an ultrasonic process on said sample.

10. The nucleic-acid extraction cartridge according to claim 9, wherein said ultrasonic-wave generation section is implemented as the same member as one of said electrodes.

11. The nucleic-acid extraction cartridge according to claim 10, wherein said damming section is a dialysis membrane or a polymer gel.

12. The nucleic-acid extraction cartridge according to claim 11, wherein said ultrasonic-wave generation section is an ultrasonic probe or an ultrasonic horn.
