Abstract

The invention relates to novel methods and compositions for the detection of analytes using the nuclear reorganization energy, λ, of an electron transfer process.

23. A method of detecting a target analyte in a test sample comprising:

a) adding a target analyte to an electrode comprising;

i) a transition metal complex with a first E⁰, said transition metal complex comprising a metal selected from the group consisting of manganese, technetium, rhenium, iron, ruthenium, osmium, cobalt, rhodium, iridium, nickel, palladium, platinum, copper, silver and gold; and

ii) a binding ligand that will bind said target analyte;

such that upon binding of said target analyte to said binding ligand the transition metal complex is formed and said first E⁰ is altered to result in a second different E⁰; and

b) measuring electron transfer of said transition metal complex at said first E⁰ and electron transfer of said transition metal complex at said second E⁰.

24. The method according to claim 23, wherein said measuring comprises applying at least a first input signal to said transition metal complex and receiving an output signal.

25. The method according to claim 23, wherein in the absence of target analyte, said first output signal does not indicate significant electron transfer at said second E⁰.

26. The method according to claim 24, wherein said first input signal comprises at least an AC component.

27. The method according to claim 24, further comprising applying said input signal at a plurality of frequencies.

28. The method according to claim 24 or 26, wherein said input signal comprises at least a DC component.

29. The method according to claim 28, further comprising applying said input signal at a plurality of voltages.

30. The method according to claim 24, wherein said output signal is a current.

31. The method according to claim 30, wherein said current is an AC current.

32. The method according to claim 23, wherein said electrode comprises gold.

33. The method according to claim 23 wherein said electrode further comprises a self-assembled monolayer.

34. The method according to claim 33, wherein said self-assembled monolayer comprises conductive oligomers.

35. The method according to claim 33, wherein said self-assembled monolayer comprises insulators.

36. The method according to claim 23, wherein said transition metal complex has at least one coordination site occupied by a polar coordination group.

37. The method according to claim 23, wherein said transition metal complex has at least one coordination site occupied by a water molecule.

38. The method according to claim 23, wherein said transition metal complex has at least two coordination sites occupied by polar coordination groups.

39. The method according to claim 23, wherein said binding ligand is covalently attached to said transition metal complex.

40. The method according to claim 23, wherein said binding ligand is covalently attached to said electrode.

41. The method according to claim 23, wherein transition metal complex is covalently attached to said electrode.

42. The method according to claim 40, wherein said covalent attachment is via a spacer.

43. The method according to claim 41, wherein said covalent attachment is via a spacer.

44. The method according to claim 42 or 43, wherein said spacer is a conductive oligomer.

45. The method according to claim 42 or 43, wherein said spacer is an insulator.
