Abstract

A method is provided for increasing the number of hematopoietic stem cells in the bone marrow, increasing the mobilization of these cells to migrate from the bone marrow to the bloodstream and elsewhere, and increasing the number of differentiating hematopoietic stem cells in the bloodstream.

Claims

1. A method for increasing the mobility of hematopoietic stem cells, the method comprising administering a composition comprising a lipopeptide or a liposaccharide to a mammal in need thereof.

4. The method of claim 1, wherein the lipopeptide is a compound of the formula:

5. The method of claim 4, wherein the peptide comprises any one of sequences SEQ ID NOS:

1-52.

6. The method of claim 5, wherein the first five amino acids of the peptide are chosen from the amino acids at the positions referred to in Table 3.

7. The method of claim 5, wherein the peptide comprises a sequence selected from the group consisting of SEQ ID NOS:

8, 16-18, 20, and 21, and substitutions thereof.

8. The method of claim 4, wherein R₁ is H and R₂ and R₃ are C₁₆ aliphatics or substitutions thereof.

9. The method of claim 4, wherein the compound is an RR or RS stereoisomer, or mixture thereof.

10. The method of claim 1, wherein the lipopeptide is a compound of the formula:

12. The method of claim 1, wherein the liposaccharide is a compound of the formula:

13. The method of claim 1, further comprising administering a radioprotectant.

14. The method of claim 13, wherein the radioprotectant is an antioxidant.

15. The method of claim 13, wherein the radioprotectant is a cytokine.

16. The method of claim 15, wherein the cytokine is a stem cell factor.

17. The method of claim 13, wherein the radioprotectant is flagellin.

21. The method of claim 1, wherein the mobility of the hematopoietic stem cells increases from the bone marrow to the blood stream.

24. A method of treating a disease comprising increasing the mobility of hematopoietic stem cells according to the method of claim 1.

The method of claim 24, wherein the disease is selected from the group consisting of cancer, an autoimmune disease, an anemia, a non-malignant hematological disorder, Wiskott-Aldrich syndrome, Chediak-Higashi syndrome, Severe Combined Immune Deficiency (SCID), thalassemia, and sickle cell anemia.

26. The method of claim 25, wherein the cancer is selected from the group consisting of leukemia, lymphoma, a malignant hematological disorder, breast cancer, testicular cancer, neuroblastoma, ovarian cancer, myeloma, and Waldenstrom'"'"'s Macroglobulinemia.

28. A method of producing an autologous stem cell population from a mammal comprising:

(a) increasing the mobility of hematopoietic stem cells according to the method of claim 1; and

(b) isolating peripheral white blood cells from the mammal,

wherein the peripheral white blood cells comprise the autologous stem cell population.

29. A method of treating an effect of a cancer treatment, the method comprising administering an autologous stem cell population produced according to the method of claim 28 to a mammal that has received a cancer treatment.
